    摘要:

    目的 探讨补肾祛湿方对多囊性卵巢综合征(PCOS)大鼠内分泌系统的影响,为治疗PCOS的有效性提供实验依据。方法 使用SD雌性大鼠灌胃来曲唑溶液(1 mg/kg),每天1次,连续灌胃21天,建立PCOS模型。将18只PCOS模型大鼠分为模型组、补肾祛湿方组和阳性组,另取6只健康大鼠作为空白组,补肾祛湿方组灌胃补肾祛湿方药液(19 g生药/kg),阳性组灌胃达英-35药液(0.42 mg/kg),空白组与模型组灌胃生理盐水,每天1次,持续 21 天。给药结束后禁食12 h,测量体重,并处死大鼠,计算单侧卵巢指数和子宫指数,ELISA法测定血清睾酮(T)、黄体生成素(LH)、胰岛素(INS)、雌二醇(E2)含量,HE染色观察卵巢组织形态,免疫组化法检测卵巢组织中胰岛素样生长因子-1(IGF-1)表达水平,RT-qPCR检测卵巢组织中细胞外调节蛋白激酶1、2(ERK1、ERK2)mRNA表达量,Western Blot检测ERK1、ERK2、磷酸化-ERK1(p-ERK1)及磷酸化-ERK2(p-ERK2)水平。结果 与空白组相比,模型组大鼠卵巢指数、子宫指数、血清中E 2 水平降低,血清中T、LH和INS水平升高,卵巢中ERK1、ERK2 mRNA、IGF-1、ERK1、ERK2、p-ERK1和p-ERK2蛋白表达水平升高(P<0.05)。与模型组相比,补肾祛湿方组和阳性组子宫指数和血清中E2水平升高、血清中T、LH和INS水平降低,卵巢中ERK1、ERK2 mRNA、IGF-1、ERK1、ERK2、p-ERK1和p-ERK2蛋白表达水平降低(P<0.05)。结论 补肾祛湿方具有改善PCOS大鼠激素紊乱和减少卵巢颗粒细胞损伤的功效,其机制可能与抑制ERK1/2蛋白表达有关

    Abstract:

    Objective This study explores the effects of Bushen Qyushi formula on the endocrine system of rats with polycystic ovary syndrome (PCOS), aiming to provide experimental evidence for the effectiveness of this treatment method in the treatment of PCOS. Methods The SD female rats were intragastrically administered the letrozole solution (1 mg/kg) once a day for 21 consecutive days to establish the PCOS model. The 18 PCOS model rats were divided into the model group, the Bushen Qyushi group and the positive control group. Another 6 healthy rats were used as the blank group. The Bushen Qyushi group was intragastrically administered Bushen Qyushi formula medicinal liquid (19 g of raw drug/kg), the positive control group was intragastrically administered the Diane-35 medicinal liquid (0.42 mg/kg), and the blank group and the model group were intragastrically administered normal saline, once a day for 21 days. After the administration, the rats were fasted for 12 hours, their body weight was measured, and then they were sacrificed. The unilateral ovarian index and uterine index were calculated. The serum testosterone (T), luteinizing hormone (LH), insulin (INS), and estradiol (E2) contents were determined by ELISA. The ovarian tissue morphology was observed by HE staining. The expression level of insulin-like growth factor-1 (IGF-1) in the ovarian tissue was detected by immunohistochemistry. The mRNA expression levels of extracellular regulated protein kinases 1 and 2 (ERK1 and ERK2) in the ovarian tissue were detected by RT-qPCR. The levels of ERK1, ERK2, phosphorylated-ERK1 (p-ERK1) and phosphorylated-ERK2 (p-ERK2) were detected by Western Blot.Results Compared with the blank group, the ovarian index, uterine index and serum E2 level in the model group decreased, while the serum T, LH and INS levels increased, and the expression levels of ERK1, ERK2 mRNA, IGF-1, ERK1, ERK2, p-ERK1 and p-ERK2 proteins in the ovary in the model group increased (P < 0.05). Compared with the model group, the uterine index and serum E2 level in Bushen Qyushi group and the positive group increased, while the serum T, LH and INS levels decreased, and the expression levels of ERK1, ERK2 mRNA, IGF-1, ERK1, ERK2, p-ERK1 and p-ERK2 proteins in the ovary in Bushen Qyushi group and the positive group decreased (P<0.05).Conclusion Bushen Qyushi formula can improve the hormone disorder and 〖JP2〗reduce the damage of ovarian granulosa cells in PCOS rats, which may be related to the inhibition of ERK1/2 protein expression
