Abstract:Objective To explore the biological behavior of LncRNA FAS-AS1 on K562 leukemia cells. Methods The K562 cell culture in the logarithmic growth phase was divided into blank group (normal culture with no treatment), negative control group (transfected with LncRNA FAS-AS1 empty plasmid), and experimental group (LncRNA FAS-AS1). And the cell proliferation was detected by MTT method, the content of mTOR and AMPK proteins was detected by Western blot, and the difference of cell apoptosis was detected by flow cytometry.Results Compared with blank control group and negative control group, LncRNA Fas-AS1 content of K562 cells in experimental group was significantly increased (all P<0.05). However, there was no significant difference in mRNA content between blank and negative control cells (0.38±0.05 vs 0.39±0.06) (P>0.05). With the extension of culture time, the proliferation rate of K562 cells in experimental group (0.540±0.023) was significantly lower than that in control group (P<0.05), but there was no significant difference between blank and negative control group (0.851±0.040 vs 0.848±0.047) (P>0.05). The apoptosis rate of the experimental group was (25.13±5.22) %, significantly higher than that of the blank control group [(14.54±1.28)%] and the negative control group [(15.35±1.30)%] (P<0.05). There was no significant difference between the blank control group and the negative control group (P>0.05). The expression of AMPK protein in K562 cells in the experimental group was significantly higher than that in the control group (P<0.05), while there was no significant difference in the protein content in K562 cells between the two groups (P>0.05).Conclusion LncRNA Fas-AS1 overexpression can significantly affect the biological behavior of K562 leukemia cells, and can reduce the number and proliferation activity of cells, which may be related to the activation of AMPK gene and the blocking of mTOR activity
