Abstract:Objective To investigate the effect of circ_0001649 on glycolysis, proliferation, migration and invasion of ovarian cancer SKOV3 cells and its possible mechanism. Methods 39 cases of ovarin cancer patients diagnosed pathologically in our hospital from February 2018 to December 2019 were selected. The expression of circ_0001649, miR-224 in ovarian cancer tissues and adjacent tissues were detected by qRT PCR. In vitro culture of human ovarian cancer cell SKOV3, pcDNA and pcDNA circ_ 0001649, anti-miR-NC, anti-miR-223, pcDNA-circ_ 0001649 and miR-NC, pcDNA circ_ 0001649 and miR-223 mimics were transfected into SKOV3 cells. The circ in SKOV3 cells_ 0001649, miR-223 expression level were observed. Glucose consumption, lactic acid, cell proliferation, migration and invasion were detected. Results Compared with adjacent tissues, the expression level of circ_0001649 in ovarian cancer tissues was decreased (P<0.05), and the expression level of miR-223 was increased (P<0.05) Transfection of pcDNA-circ_0001649 or transfection of anti-miR-223 could significantly reduce glucose consumption and lactate level (P<0.05), and reduce OD value and the protein levels of Ki-67, MMP-2 and MMP-9 (P<0.05), reduce the number of migration and invasion cells (P<0.05). The dual luciferase report experiment confirmed that circ_0001649 could target miR-223. Co-transfection of pcDNA-circ_0001649 and miR-223 mimics could significantly increase the OD value and the protein levels of Ki-67, MMP-2, and MMP-9 (P<0.05), increase glucose consumption and lactate levels (P<0.05), and increase the number of migration and invasion cells (P<0.05).Conclusion circ0001649 overexpression can down regulate the expression of miR-223, thereby inhibiting the development of ovarian cancer SKOV3 cells
