Abstract:Objective To investigate the potential role of Mfn2 and miRNA-101 in regulating hepatic fibrosis in hepatic alveolus echinococcosis through TGF-β1. Methods Liver tissues removed after surgery were collected from 20 patients with hepatic alveolar echinococcosis diagnosed in Qinghai Provincial People's Hospital from July 2020 to October 2020. Normal liver tissues more than 2cm away from the edge of liver lesions and liver tissues less than 2cm away from the edge of liver lesions were collected. HE and Masson staining were performed to grade fibrosis of different specimens. The expressions of Mfn2, TGF-β1, α-SMA and miRNA-101 in different grades of fibrosis were detected by immunohistochemistry and RT-qPCR. Results The expressions of Mfn2, TGF-β1, α-SMA and miRNA-101 in different grades of fibrosis were statistically significant (P<0.05). The expression of Mfn2 was negatively correlated with TGF-β1 and α-SMA (P<0.05). The expression levels of miRNA-101 were negatively correlated with TGF-β1 and α-SMA (P<0.05),TGF-β1 was positively correlated with α-SMA (P<0.05), and Mfn2 was not significantly correlated with miRNA-101 (P>0.05). Conclusion Mfn2 and miRNA-101 may inhibit hepatic fibrosis in hepatic alveolar echinococcosis, and may play an antifibrotic role by inhibiting TGF-β1
