    摘要:

    目的 探讨二甲双胍在胃癌细胞的增殖、迁移及上皮间质转化之间的作用。方法 用不同浓度(0、10、20、40 mmol/L)的二甲双胍处理胃癌细胞,将细胞分为对照组、二甲双胍处理组(10、20、40 mmol/L)。采用CCK8试剂检测不同浓度二甲双胍处理的胃癌细胞的增殖活性。划痕实验及Transwell实验检测二甲双胍对胃癌细胞迁移和侵袭的影响。采用Western blot 检测上皮-间充质转化(EMT)相关蛋白水平。结果 CCK8检测结果显示,与对照组比较,二甲双胍处理组随浓度增加增殖活性显著降低,呈浓度依赖(F=22.65,P=0.0003)。划痕实验结果显示,与对照组比较,二甲双胍处理组随浓度增加迁移率显著降低,呈浓度依赖(F=27.22,P<0.0001)。Western blot结果显示,二甲双胍处理组E-cadherin蛋白水平均高于对照组(F=64.82,P<0.0001),vimentin蛋白水平均低于对照组(F=11.28,P=0.003)。结论 二甲双胍能够抑制胃癌细胞增殖、迁移,其在胃癌转移中发挥抑制作用,可能与抑制上皮-间充质转化相关。

    Abstract:

    Objective To investigate the effects of metformin on the proliferation, migration and epithelial mesenchymal transition of gastric cancer cells.Methods Gastric cancer cells were treated with different concentrations of metformin (0,10, 20, 40 mmol/L). And the cells were divided into four groups: CN group and metformin treated group (10, 20, 40 mmol/L). Cck8 reagent was used to detect the proliferation activity of GC cells treated with different concentrations of metformin. Scratch test and Transwell test detect the influence of metformin on migration in GC cells. Western blot was used to detect changes in the expression level of EMT (epithelial-mesenchymal transition) related proteins in GC cells.Results Compared with the control group, the proliferation activity of the experimental group decreased significantly with the increase of concentration (F=22.65).Compared with the control group, the migration rate of the experimental group decreased significantly with the increase of concentration (F=27.22, P=0.0003), Western blot results showed that the expression of E-cadherin protein in experimental group was higher than that in control group, and vimentin protein expression was lower than that in control group (E-cadherin protein:F=64.82, P<0.0001;vimentin protein:F=11.28, P=0.003).Conclusion Metformin inhibits the ability of migration by inhibiting EMT in GC cells.
