Abstract:Objective To explore the effect of honeycomb extract Elixir-X on natural aging and radiation aging mice, and provide a pharmacological basis for the further development of Elixir-X. Methods Mice were divided into blank control group, Elixir x group, resveratrol group and model group. The changes of senescent cells, liver function recovery and liver regeneration during regeneration after 70% liver resection in mice were detected, to observe the effect of Elixir-X on natural senescence. The mice model of radiation aging was established using co-60 coefficient CO radiation method and divided into model group, blank control group, resveratrol group, high dose and low dose Elixir-X group. The changes of intestinal crypt stem cell regeneration, villus structure and senescent cells were detected. The effects of different doses of Elixir-X on senescence after radiation were compared. Results In hepatectomy mice, the liver function level of Elxir-X treatment group was significantly lower than that of normal liver regeneration mice, and significantly promoted liver function recovery (P<0.05). Compared with the model group, the senescent cells in the Elixir-X group were removed (P<0.05), and the staining plaque of Ki-67 was significantly increased, which promoted the regeneration of liver. Compared with model group, Pressume 60 Species in mice irradiated with Elixir-X can effectively promote liver function recovery (P<0.05), and body weight of Elixir-X group recovered to normal level, no significant difference was found between the two groups (P>0.05). Compared with model group, β-Gal staining cells were decreased in Elixir-X-L group, and even decreased senescent cells more effectively in Elixir-X-H group (P<0.05). The small intestinal villi were recovered in the treatment group of Elixir-X by HE staining. In IHC staining of Ki-67, there were more staining plates in the Elixir-X group compared with the model group, and the staining areas were all in the position of intestinal crypt stem cells. Conclusion Elixir X can effectively eliminate the natural aging caused by liver resection and the damaged senescent cells caused by radiation, and it has a significant inhibition effect on aging.
