    摘要:

    目的 观察内质网应激反应(ERS)对哮喘大鼠呼吸道黏膜上皮细胞黏蛋白5AC(MUC5AC)的作用。方法 50只大鼠随机分为对照组、卵清白蛋白(OVA)组、小干扰RNA(siRNA)组、阴性对照(NC)组及4-苯基丁酸(4-PBA)组,每组10只。除对照组外,其余组采用OVA建立哮喘模型。相应干预后,比较各组Penh值;计算肺泡灌洗液(BALF)中性粒细胞、嗜酸性粒细胞、淋巴细胞数百分比(%);检测BALF中白介素(IL)-4、干扰素(IFN)-γ水平;观察肺组织病理变化;检测Sec61、葡萄糖调节蛋白78(GRP78)、C/EBP环磷酸腺苷反应元件结合转录因子同源蛋白(Chop)、MUC5AC蛋白表达。结果 与对照组比较,OVA组Penh、BALF中细胞总数、中性粒细胞、嗜酸性粒细胞数和淋巴细胞百分比,IL-4,Sec61、GRP78、Chop、MUC5AC升高,IFN-γ降低(均P<0.05);与OVA组比较,siRNA组Penh、BALF中细胞总数、中性粒细胞、嗜酸性粒细胞数和淋巴细胞百分比,IL-4,Sec61、GRP78、Chop、MUC5AC降低,IFN-γ升高(均P<0.05)。HE染色显示,OVA组杯状细胞增生,支气管管壁增厚,可见炎性细胞浸润;siRNA组较OVA组病理有所改善。结论 Sec61可能通过调节ERS,影响MUC5AC表达,参与哮喘发生、发展过程。

    Abstract:

    Objective To observe the mechanism of endoplasmic reticul stress (ERS) on the expression of mucin 5AC (MUC5AC) in respiratory mucosal epithelial cells of asthmatic rats. Methods Fifty rats were randomly divided into control group, ovalbumin (OVA) group, small interfering RNA (siRNA) group, negative control (NC) group and 4 phenylbutyric acid (4-PBA) group with 10 rats in each group. In addition to the control group, the other groups were established asthma model by ova. After corresponding intervention, Penh value of each group was compared; The percentagesof neutrophils, eosinophils and lymphocytes in BALF were calculated; IL-4 and IFN-4 were detected in BALF-γ Level; The pathological changes of lung tissue were observed; Sec61, GRP78, chop and MUC5AC were detected. Results Compared with the control group, the total number of cells, neutrophils, eosinophils, percentage of lymphocytes, IL-4, Sec61, GRP78, chop and MUC5AC in Penh and BALF of ova group were increased, while IFN 4, Sec61, GRP78, chop and MUC5AC were increased γ0 Compared with OVA group, the total number of cells, neutrophils, eosinophils, percentage of lymphocytes, IL-4, Sec61, GRP78, chop and MUC5AC in Penh and BALF of siRNA group were lower than those of ova group γ0 He staining showed that goblet cells proliferated, bronchial wall thickened and inflammatory cells infiltrated in ova group. The pathology of siRNA group was better than that of ova group.Conclusion Sec61 may affect the expression of MUC5AC by regulating ERS, and participate in the occurrence and development of asthma.
