    【摘要】目的 探讨茯苓酸(PA)对过氧化氢(H2O2)诱导的脑微血管内皮细胞凋亡及增殖的影响。方法 体外培养小鼠脑微血管内皮细胞株bEnd.3,随机分为NC组(正常培养细胞)、H2O2组(H2O2浓度为100 μmol/L)、H2O2+PA 10 μmol/L组(H2O2浓度为100 μmol/L,PA浓度为10 μmol/L)、H2O2+PA 20 μmol/L组(H2O2浓度为100 μmol/L,PA浓度为20 μmol/L)、H2O2+PA 40 μmol/L组(H2O2浓度为100 μmol/L,PA浓度为40 μmol/L)。采用甲基噻唑基四唑(MTT)法检测细胞存活率;流式细胞术检测细胞凋亡率;检测乳酸脱氢酶(LDH)、丙二醛(MDA)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSHPx)、活性氧(ROS)的水平;蛋白免疫印迹法(Western blot)检测活化的含半胱氨酸的天冬氨酸蛋白水解酶3(Cleaved caspase3)的表达量。结果 与NC组比较,H2O2组细胞存活率及CAT、SOD、GSHPx的水平显著降低(P<005),LDH、MDA、ROS的水平显著升高(P<005),细胞凋亡率与Cleaved caspase3蛋白水平显著升高(P<005);与H2O2组比较,H2O2+PA 10 μmol/L组、H2O2+PA 20 μmol/L组、H2O2+PA 40 μmol/L组细胞存活率及CAT、SOD、GSHPx的水平显著升高(P<0.05),LDH、MDA、ROS的水平显著降低(P<005),细胞凋亡率与Cleaved caspase3蛋白水平显著降低(P<005)。结论 茯苓酸可抑制H2O2诱导的脑微血管内皮细胞凋亡及促进细胞存活。

    Abstract:

    【Abstract】Objective To explore the effect of PA on the apoptosis and proliferation of brain microvascular endothelial cells induced by H2O2. Methods The mouse brain microvascular endothelial cell line bEnd.3 was cultured in vitro and randomly divided into NC group (normally cultured cells), H2O2 group (H2O2 concentration 100 μmol/L), H2O2+PA 10 μmol/L group (H2O2 concentration 100 μmol/L, PA concentration 10 μmol/L), H2O2+PA 20 μmol/L group (H2O2 concentration 100 μmol/L, PA concentration 20 μmol/L), H2O2 + PA 40 μmol/L group (H2O2 concentration 100 μmol/L, PA concentration 40 μmol/L). MTT method was used to detect cell survival rate. Flow cytometry was used to detect the apoptosis rate. The levels of LDH, MDA, CAT, SOD, GSHPx, ROS were detected. Western blot was used to detect the expression of Cleared caspase3. Results Compared with the NC group, the cell survival rate and the levels of CAT, SOD and GSHPx in the H2O2 group were significantly reduced (P <005), and the levels of LDH, MDA and ROS were significantly increased (P <005), the level of caspase3 protein was increased significantly (P<005). Compared with the H2O2 group, the cell survival rate and the levels of CAT, SOD, GSHPx in H2O2+PA 10 μmol/L group, H2O2+PA 20 μmol/L group, H2O2+PA 40 μmol/L group were significantly increased (P<005), the levels of LDH, MDA, and ROS were significantly reduced (P<005), and the apoptosis rate and the level of Cleared caspase3 protein were significantly reduced (P<005). Conclusion Poriac acid could inhibit H2O2induced apoptosis of cerebral microvascular endothelial cells and promote cell survival.
