Abstract:【Abstract】Objective To study the effects of Miao medicine Xuemaitong on autophagy and apoptosis of brain tissue in rat models of cerebral hemorrhage based on VEGF signaling pathway and its corresponding role mechanism. Methods Rat models of cerebral hemorrhage were established according to the Rosenberg method. Rats were randomly divided into sham operation group, cerebral hemorrhage group, administration group A and administration group B. Administration group A and administration group B were treated with 15 mg/(kg·d) and 30 mg/(kg·d) of Miao medicine Xuemaitong respectively. The sham operation group and cerebral hemorrhage group were given the same amount of normal saline, and they were given for 35 d. The apoptosis of each group was detected by TUNEL method. The expression of Beclin-1, VEGF, stromal cell-derived factor 1 (SCDF-1α) and CXCR-4 protein was detected by Western blot. The protein levels of PI3K andAKT were detected by immunohistochemistry. The volume of hemorrhagic lesions was measured in each group. Results The quantity of apoptotic cells and the volume of hemorrhagic lesions in cerebral hemorrhage group, administration group A and administration group B were significantly higher than those in sham operation group B, and the protein expression levels of Beclin-1, VEGF, PI3K, AKT, stromal cell-derived factor 1 and CXCR-4 were significantly lower than those in sham operation group. Compared with cerebral hemorrhage group, the quantity of apoptotic cells and the volume of hemorrhagic lesions in administration group A and administration group B were significantly decreased while the protein expression levels of Beclin-1, VEGF, PI3K, AKT, stromal cell-derived factor 1 and CXCR-4 were significantly increased (P<0.05). Conclusion Miao medicine Xuemaitong can treat brain cerebral hemorrhage by activating VEGF signaling pathway, regulating cell autophagy, inhibiting apoptosis and exerting brain protection.
