Abstract:【Abstract】 Objective To investigate the effect of lncRNA LINC00987 on proliferation, invasion and migration of lung cancer cells. Methods Taking normal alveolar epithelial cell HPAEPIC as the control, the expression levels of LINC00987 in lung cancer cell lines A549, HCC827 and H1299 were detected by qRT-PCR. A549 cells are divided into pcDNA-control group, pcDNA-LINC00987 group, LINC00987(WT)+miR-control group, LINC00987(WT)+miR-375 group, LINC00987(MUT)+miR-control group, LINC00987(MUT)+miR-375 group, si-control group, si-LINC00987 group, miR-control group, miR-375 group, anti-miR-control group, anti-miR-375 group, pcDNA-LINC00987+miR-control group and pcDNA-LINC00987 +miR-375 group. The proliferation, invasion and migration abilities of A549 cells were determined by CCK8 and Transwell assay, respectively. The relationship between LINC00987 and miR-375 was verified by dualluciferase reporter assay system. Results Compared with normal alveolar epithelial cells HPAEPIC, the expression levels of LINC00987in lung cancercell lines A549, HCC827 and H1299were decreased significantly (P<0.05), and the following experiments were carried out on A549 cells with remarkably different expression levels. pcDNA-LINC00987 group inhibited the proliferation, invasion and migration of A549 cells. LINC00987 targeted and inhibited the expression of miR-375. The miR-375 group could promote the proliferation, invasion and migration of A549 cells, while the anti-miR-375 group had the opposite result; the pcDNA-LINC00987+miR-375 group promoted the proliferation, invasion and migration of A549 cells. Conclusion LncRNA LINC00987 inhibits proliferation, invasion and migration of lung cancer A549 cells by targeting miR-375. LINC00987 may be a potential molecular target for lung cancer.
