Abstract:Objective To explore the antitumor effect and mechanism of Guizhi Fuling pill on rat ovarian cancer xenografts. Methods 30 SD male rats were randomly divided into control group, model group and Guizhi Fuling pill group. Model group and Guizhi Fuling pill group were subcutaneously inoculated with human ovarian cancer HO8910 cells to establish a rat ovarian cancer xenograft model. After successful modeling, Guizhi Fuling pill group was intragastrically administered with Guizhi Fuling pill for 10 d, and control group and model group were given equal volume of distilled water. After treatment, the volume and weight of ovarian cancer xenografts were detected, and the levels of serum follicle stimulating hormone(FSH), luteinizing hormone(LH), serum carbohydrate antigen(CA) 125, interleukin(IL)2 and IL6 were measured. Fluorogenic quantitative PCR and Western Blot were used to detect the protein and mRNA expression levels of caspase-3, survivin and p21waf/cip in ovarian cancer xenografts. Results Compared with model group, the volume and weight of ovarian cancer xenografts were significantly decreased in Guizhi Fuling pill group (P<0.05). The serum IL-2 level was significantly increased (P<0.05). The levels of serum FSH, LH, CA125 and IL-6 were significantly decreased P<0.05),and the protein and mRNA expression levels of survivin were significantly decreased(P<0.05). The protein and mRNA expression levels of Caspase-3 and p21waf/cip were significantly increased (P<0.05). Conclusion Guizhi Fuling pill can enhance the immunity of body and promote the apoptosis of tumor cells, thus achieving antitumor effects.
