Abstract:Objective To investigate the impact of ischemia reperfusion injury on the TNFα/IL-6 /STAT-3 pathway in the remnant liver after subtotal hepatectomy models. Methods 60 male SD rats weighting 230~280g were obtained and randomly divided into groupⅠ (subtotal hepatectomy, SH), group Ⅱ (ischemia reperfusion, IR). The left and middle liver was resected, and the remnant liver was treated as followed: the blood flow was not blocked in SH group, but blocked 30 minutes in IR group, then reperfused. After 1, 6, 12, 24 and 48h, respectively, the serum TNFα, IL-6 was detected. The mRNA of CyclinD1, Cdk4 and STAT3 were assayed. The protein expression of CyclinD1 and Cdk4 were detected. Results Compared with SH group, the expression of IL6 declined at each indicated time point, but the content of TNFα increased in early stage(1~12h) in IR group. Moreover, the expression of CyclinD1, Cdk4 and STAT3 was significant decrease in IR group than that in SH group in 24h and 48h. Conclusion Ischemia reperfusion injury could reduce the expression of TNF/il-6 /STAT-3 signaling pathway in the residual liver tissue after subtotal hepatectomy, and may ultimately inhibit the proliferation of hepatocytes.
