Abstract:【Abstract】 Objective To construct a RNA regulatory network for colorectal cancer and explore the molecular regulation mechanism of colorectal cancer. Methods The transcriptional level expression data of colorectal cancer were extracted from TCGA database, differentially expressed RNAs was selected. According to target prediction databases, miRDB, miRcode, miRTarBase and TargetScan, RNA pairs of Lnc-RNA-miRNA and miRNA-mRNA were identified. Basing on ceRNA hypothesis, and analysis of LncRNA intracellular localization, the cytoplasmic expression LncRNAs were identified to regulate the network. Finally the InRNA-miRNA-mRNA in colorectal cancer was reconstructed. Results A total of 119 LncRNA, 32 miRNAs and 59 mRNAs were screened from differentially expressed RNAs in colorectal cancer. 777 pairs of LncRNA-miRNA and 77 pairs of miRNA-mRNA were identified. Based on the ceRNA hypothesis and cytoplasm expressed LncRNAs, 15 up-regulated LncRNAs, 3 down-regulated miRNAs (hsa-mir-145, hsa-mir-193b, hsa-mir-150) and 5 up-regulated mRNAs were selected to reconstruct the ceRNA network. Conclusion The RNA network mediated by LncRNA in colorectal cancer was successfully constructed, and the potential targets were identified, which provided a reference for the treatment of colorectal cancer.
