海昆肾喜胶囊对慢性肾衰竭大鼠肾功能的保护作用

    【摘要】 目的 探讨海昆肾喜胶囊对慢性肾衰竭(CRF)大鼠肾功能病理状态的治疗作用。方法 在45只清洁级大鼠中选取10只作为对照组,其余35只仅25只造模成功,将造模成功大鼠随机分为模型组(12只)和实验组(13只),模型组和实验组采用腺嘌呤建立CRF模型。实验组给予海昆肾喜胶囊溶液(011g/100g·d)灌胃,对照组与模型组动物给予等量生理盐水灌胃。给药4周后,测定肾功能相关指标血清尿素氮(BUN)、肌酐(Scr)水平、机体炎症因子、肿瘤坏死因子α(TNFα)、白介素6(IL6)水平、肾组织中纤维连接蛋白(FN)、层黏链蛋白(LN)、α平滑肌肌动蛋白(αSMA)和转化生长因子β(TGFβ)蛋白表达情况,采用Masson染色法测定肾组织纤维化情况。结果 与对照组相比,模型组和实验组中BUN、Scr、TNFα和IL6含量显著升高(P<005),αSMA和TGFβ蛋白表达量明显增多(P<005);与模型组相比,实验组中BUN、Scr、TNFα和IL6含量显著降低(P<005),αSMA和TGFβ蛋白表达明显减少(P<005),Masson染色中实验组肾组织纤维化情况明显改善;免疫组化实验中,实验组FN与LN蛋白表达明显减小。结论 海昆肾喜胶囊可通过抑制肾组织纤维化进程、降低炎症因子分泌,从而对大鼠慢性肾衰竭状态起到治疗作用。

    【Abstract】 Objective To investigate the treatment effects of Haikun Shenxi capsule on renal function and pathological state in rats with chronic renal failure. Methods 35 rats were randomly divided into control group, model group and experimental group. Model group and experimental group were given adenine to establish CRF model. After successful modeling, the experimental group was intragastrically administered with Haikun Shenxi capsule solution (011g/100g·d), and control group and model group were given the same amount of normal saline. The renal functionrelated indexes of blood urea nitrogen (BUN) and creatinine (Scr) and inflammatory factors of tumor necrosis factorα (TNFα) and interleukin6 (IL6) were measured. The expression levels of fibronectin (FN), laminin (LN), αsmooth muscle actin (αSMA) and transforming growth factorβ (TGFβ) in renal tissues were measured. The renal tissue fibrosis was determined by Masson staining. Results Compared with control group, the contents of BUN, Scr, TNFα and IL6 in model group and experimental group were significantly increased (P<005), and the protein expression levels of αSMA and TGFβ were significantly increased (P<005). Compared with model group, the contents of BUN, Scr, TNFα and IL6 in experimental group were significantly decreased (P<005), and the protein expression levels of αSMA and TGFβ were significantly reduced (P<005). The renal tissue fibrosis in experimental group was significantly improved in Masson staining. In the immunohistochemistry experiment, the protein expression levels of FN and LN in experimental group were significantly reduced. Conclusion Haikun Shenxi capsule can treat the chronic renal failure in rats by inhibiting the process of renal tissue fibrosis and reducing the secretion of inflammatory factors.
