    【摘要】 目的 探讨梓醇通过调控miR200表达抑制卵巢癌细胞增殖和促进其凋亡的行为及其机制。方法 采用定量多聚酶琏反应(qPCR)检测不同浓度的梓醇对卵巢癌细胞的细胞活性影响情况,分析梓醇对卵巢癌细胞增殖行为的影响情况;2,5一二本基四唑溴化物(MTT)实验检测梓醇对卵巢癌细胞增殖能力的影响,qPCR检测梓醇对半胱天蛋白3表达水平的影响,流式细胞术实验检测梓醇对卵巢癌细胞凋亡行为时间依赖性的的影响;Western blotting实验检测梓醇对卵巢癌细胞中基质金属蛋白酶2(MMP2)蛋白表达情况的影响及miR200对其的逆转作用。结果 100μg/ml的梓醇对卵巢癌细胞的细胞活性的抑制作用最佳;而随着时间的进展,梓醇对卵巢癌细胞的增殖能力的直接抑制作用,可以在一定程度上干扰卵巢癌细胞的生长进程;而合适浓度梓醇对卵巢癌细胞中MMP2蛋白有直接的抑制作用,转染miR200inhibitor后可以在一定程度上逆转梓醇对卵巢癌细胞增殖行为的作用。结论 梓醇可以通过调控miR200表达抑制卵巢癌细胞的增殖和促进其凋亡行为,可为临床治疗和预后提供参考。

    Abstract:

    【Abstract】 Objective To investigate the inhibitory effect and mechanism of catalpol on the proliferation and apoptosis of ovarian cancer cells by regulating miR200 expression. Methods qPCR was used to detect the effect of different concentrations of catalpol on the cell viability of ovarian cancer cells. The effect of catalpol on the proliferation of ovarian cancer cells was analyzed. The effect of catalpol on the proliferation of ovarian cancer cells was detected by MTT assay. The effect of catalpol on the expression of caspase3 was detected by qPCR. Flow cytometry was used to detect the effect of catalpol on the apoptosis of ovarian cancer cells in a timedependent manner. Western blotting was used to detect the effect of catalpol on the expression of MMP2 in ovarian cancer cells. Results 100μg / ml catalpol had the best inhibitory effect on the cell activity of ovarian cancer cells. With the progress of time, catalpol directly inhibits the proliferation of ovarian cancer cells can interfere with the growth of ovarian cancer cells to a certain extent. Appropriate concentration of catalpol had a direct inhibitory effect on MMP2 protein in ovarian cancer cells. The transfection of miR200inhibitor can reverse the effect of catalpol on the proliferation of ovarian cancer cells to a certain extent.Conclusion Catalpol can inhibit the proliferation and promote the apoptosis of ovarian cancer cells by regulating the expression of miR200.
