预适应联合后适应对大鼠缺血再灌注损伤的影响

    摘要:

    目的 探讨预适应联合后适应对大鼠脑缺血再灌注损伤的影响及保护作用。方法 选取清洁级雄性SD大鼠50只,随机分为假手术组、模型组、预适应组、后适应组和预适应+后适应组,每组10只,除假手术组外,其余各组大鼠均制作为脑缺血/再灌注损伤模型,在再灌注48h后,检测各组大鼠神经功能、脑梗死灶体检、脑组织氧化应激损伤生化指标及神经元凋亡情况。结果 再灌注12、24和48h模型组神经功能缺陷评分明显高于其他处理组(P<0.05);再灌注12、24和48h处理组中,预适应+后适应组神经功能缺陷评分最低(P<0.05);预适应+后适应组脑梗死灶体积为(100.41±33.26)mm3,明显低于模型组、预适应组和后适应组(P<0.05);预适应组和后适应组脑梗死灶体积较模型组降低(P<0.05);假手术组超氧化物歧化酶(SOD)活性明显高于模型组,而丙二醛(MDA)低于模型组(P<0.05);;后适应组和预适应+后适应组SOD活性较模型组升高,而MDA较模型组降低(均P<0.05);假手术组细胞凋亡数明显低于模型组(P<0.05);各处理组细胞凋亡数较模型组有所降低(P<0.05),其中预适应+后适应组神经元细胞凋亡数最少,为(47.20±9.21)个。结论 预适应和后适应均能对大鼠脑缺血再灌注损伤具有保护作用,而两者联合应用能进一步加强保护作用。

    Abstract:

    Objective To investigate protective effects of preconditioning combined with post adaptation on cerebral ischemia / reperfusion injury in rats. Methods 50 clean grade male SD rats were randomly divided into sham operation group, model group, pre adaptation group, post adaptation group and preconditioning + post adaptation group, with 10 rats in each group. except sham operation group, the rats were made into cerebral ischemia / reperfusion injury model. After reperfusion 48h, the neurological function, cerebral infarction, brain tissue oxidative stress injury biochemical indicators and neuronal apoptosis in rats were observed. Results The nerve function defect score of model group after reperfusion 12, 24 and 48h was significantly higher than that of other groups (P<0.05). The nerve function defect score of preconditioning + post adaptation group after reperfusion 12, 24 and 48h was the lowest among the groups (P<0.05). The infarct volume of preconditioning + post adaptation group (100.41±33.26 mm3) was significantly lower than that of model group, preconditioning group and post adaptation group (P<0.05). The volume of cerebral infarction in the preconditioning group and the post adaptation group were lower than that of the model group (P<0.05). The superoxide dismutase (SOD) activity of Sham operation group was significantly higher than that in the model group (P<005). The malondialdehyde (MDA) activity of Sham operation group was significantly lower than that in the model group (P<0.05). The SOD activity of post adaptation group and preconditioning + post adaptation group were higher than that in the model group, but MDA of post adaptation group and preconditioning + post adaptation group were lower than that in the model group (P<005). The number of apoptotic cells of Sham operation group was significantly lower than that in the model group (P<0.05). Conclusion Preconditioning and post adaptation can protect the cerebral ischemia / reperfusion injury in rats, and the combined application can further strengthen the protective effect.
