    目的 观察脂肪和肥胖相关蛋白(FTO)在非小细胞肺癌(NSCLC)中的表达及其对非小细胞肺癌(NSCLC)血管生成的作用并探讨机制。方法 RT-qPCR检测45个NSCLC组织和癌旁组织中FTO的表达水平,蛋白免疫印迹观察组织中FTO的蛋白表达,分为癌旁和NSCLC组。使用shRNA敲低FTO,分为sh-NC和sh-FTO组。RT-qPCR检测5个肺癌细胞系和气道上皮细胞中FTO的表达水平,蛋白免疫印迹观察细胞中FTO的蛋白表达。细胞计数试剂盒8(CCK-8)和荷瘤实验分别检测FTO对NSCLC细胞在体外和体内增殖能力的影响。Tube形成实验观察小管形成的长度。结果 FTO在45例NSCLC组织中的mRNA和蛋白表达高于癌旁组织(均P<0.05)。FTO的体外肺癌细胞表达同样高于气道上皮细胞(均P<0.05)。敲低FTO(sh-FTO)抑制NSCLC细胞在体内和体外的增殖(均P<0.05),且敲低FTO抑制NSCLC细胞的小管形成长度(P<0.05)。结论 FTO在NSCLC组织中的表达高于癌旁组织。低表达FTO通过抑制增殖和血管新生影响NSCLC的发生和发展。

    Abstract:

    Objective To observe the expression of obesity-related protein (FTO) in non-small cell lung cancer (NSCLC) and its effect on NSCLC angiogenesis and explore the mechanism.Methods RT-qPCR was used to detect the expression level of FTO in 45 NSCLC tissues and adjacent tissues. Western blotting was used to observe the protein expression of FTO in the tissues. They were divided into adjacent and NSCLC groups. ShRNA was used to knock down FTO in vitro, and the cells were divided into sh-NC and sh-FTO groups. RT-qPCR was used to detect the expression level of FTO in 5 lung cancer cell lines and airway epithelial cells, and the protein expression of FTO in the cells was observed by western blotting. Cell counting kit 8 (CCK-8) and tumor-bearing experiment were used to detect the effects of FTO on the proliferation of NSCLC cells in vitro and in vivo. The tube formation experiment observes the length of the tube formation.Results The mRNA and protein expression of FTO in 45 cases of NSCLC tissues was higher than that of adjacent tissues (all P<0.05). The expression of FTO in lung cancer cells in vitro was also higher than that of airway epithelial cells (all P<0.05). Knockdown of FTO (sh-FTO) inhibited the proliferation of NSCLC cells in vivo and in vitro (both P<0.05), and knockdown of FTO inhibited the tubule formation length of NSCLC cells (P<0.05).Conclusion The expression of FTO in NSCLC tissue is higher than that in adjacent tissues. Low expression of FTO affects the occurrence and development of NSCLC by inhibiting proliferation and angiogenesis.
