    目的 研究白藜芦醇调控miR-506对黑色素瘤细胞增殖、迁移、侵袭的影响和机制。方法 qRT-PCR测定白藜芦醇处理后黑色素瘤细胞中miR-506表达变化,在黑色素瘤细胞中转染miR-506 inhibitor,qRT-PCR测定干扰效果。用白藜芦醇处理下调miR-506的黑色素瘤细胞,MTT测定细胞增殖,克隆形成实验测定细胞克隆能力,划痕愈合实验测定迁移,Transwell小室测定细胞侵袭,Western blot测定细胞中MMP-2和MMP-9蛋白表达变化。结果 白藜芦醇处理后的黑色素瘤细胞中miR-506表达水平上调。miR-506 inhibitor可明显抑制细胞中miR-506的表达。白藜芦醇可以明显下调黑色素瘤细胞的增殖、克隆、迁移和侵袭能力。与单纯白藜芦醇处理的细胞比较,下调miR-506后的黑色素瘤细胞经白藜芦醇处理后细胞增殖、克隆、侵袭、迁移和MMP-2、MMP-9蛋白表达水平均明显升高。结论 白藜芦醇通过上调miR-506抑制黑色素瘤细胞增殖、迁移和侵袭。

    Abstract:

    Objective To study the effect and mechanism of resveratrol on proliferation, migration and invasion of melanoma cells. Methods qRT-PCR was used to detect the expression of miR506 in melanoma cells after resveratrol treatment. miR506 inhibitor was transfected into melanoma cells and qRTPCR was used to detect the interference effect. Resveratrol reduced miR506 melanoma cells, MTT assay for cell proliferation. Colony forming assay was used to determine cell clone ability. Scratch healing test was used to determine migration. Cell invasion was measured in Transwell chamber. Western blot was used to detect the expression of MMP-2 and MMP-9 protein in cells. Results The expression level of miR-506 in resveratrol treated melanoma cells was upregulated. miR506 inhibitor significantly inhibited the expression of miR506 in cells. Resveratrol can significantly reduce the proliferation, clone, migration and invasion of melanoma cells. Compared with cells treated with resveratrol alone, the expression of MMP-2, MMP-9 and proliferation, cloning, invasion, migration of melanoma cells treated with resveratrol were significantly increased after downregulation of miR-506. Conclusion Resveratrol inhibits proliferation, migration and invasion of melanoma cells by up regulation of miR-506.
