苦瓜总皂苷对阿霉素肾病大鼠肾脏组织Smad3和Smad7 表达的影响研究

    【摘要】目的 探讨苦瓜总皂苷对阿霉素肾病大鼠肾脏组织Smad3蛋白和Smad7蛋白表达的影响,为我国中草药的开发提供参考。方法 50只雄性SD大鼠随机分为正常组、模型组及苦瓜总皂苷低、中和高剂量治疗组。除正常组外,其余组大鼠静脉注射阿霉素62 mg/kg造模。造模成功后,正常组和模型组给予生理盐水灌胃,苦瓜总皂苷低、中和高剂量治疗组分别给予苦瓜总皂苷10mg/(kg·d),20mg/(kg·d),40mg/(kg·d)剂量灌胃。8 周后处死大鼠,收集血及肾脏组织,检测24小时蛋白尿、血尿素氮 (Bun)以及血肌酐(Cr)含量。HE染色检测其肾脏组织病理学改变,免疫组化、Western blot以及RTPCR检测肾脏组织中Smad3和Smad7的表达。结果 与正常组比较,模型组和苦瓜总皂苷治疗组24小时尿蛋白在治疗8周后明显升高(P<0.05),而苦瓜总皂苷3个治疗组相较于模型组有明显降低(P<0.05)。苦瓜总皂苷治疗能明显降低阿霉素肾病大鼠的Bun和Cr。HE染色结果显示,与正常组比较,模型组出现明显的肾小球硬化同时伴有明显的肾细胞损伤,苦瓜总皂苷治疗后,上述症状均有明显的改善。免疫组化、Westernblot以及RTPCR结果显示,与正常组相比,模型组Smad3明显升高(P<0.05),Smad7明显下降(P<0.05);苦瓜总皂苷治疗后,Smad3表达明显下降(P<0.05),Smad7蛋白表达升高(P<0.05)。结论苦瓜总皂苷能明显改善糖尿病肾病已经受损的肾小球以及肾小管损伤,同时降低24小时蛋白尿,调节Bun和Cr,这可能与其可以调节Smad3和Smad7蛋白表达有关。

    【Abstract】Objective To investigate the effect of total saponins of momordicq charantia on Smad3 protein and Smad7 protein expression in rats with adriamycininduced nephropathy, and provide reference for the development of Chinese herbal medicine. Methods Fifty male SpragueDawley rats were randomly divided into normal group, model group and total saponins group. Except the control group, other groups were received intravenous injection of doxorubicin 62 mg/kg. After the success built the model, the normal group and the model group were given normal saline, and the total saponins of momordica charomdia were given 10mg/kg /d, 20mg/kg/d, 40mg/kg/d, 8 weeks. The blood useanitrogen (Bun) and serum creatinine (Cr) were measured. The renal tissues were detected by HE staining. The expression of Smad3 and Smad7 were detected with Western blot, RTPCR and Immunohistochemistry. Results Compared with the normal group, after 8 weeks of treatment, 24 hours urine protein of the model group and the total saponins groups were significantly increased compared with the model group. 24 hours urine protein of the three treatment groups were significantly lower than that of the model group. The total saponins can significantly reduce the Bun and Cr. Compared with the normal group, the model group and total saponins group showed significant glomerular sclerosis accompanied by significant renal cell injury (P<0.05). Compared with the normal group, Smad3 was significantly increased in the model group (P<0.05), and the levels of Smad7 were significantly decreased (P<0.05). The expression of Smad7 protein was significantly increased (P<0.05). Conclusion The total saponins of momordica charantia can significantly improve the glomeruli and renal tubular injury, decrease the proteinuria for 24 hours and regulate Bun and Cr, which can regulated the Smad3 and Smad7 protein.
