Abstract:【Abstract】 Objective To study the abnormal expressions of long noncoding RNA PVT1 (LncRNA PVT1) and micro RNA30d (miR30d) in epithelial ovarian cancer, and explore the relationships between LncRNA PVT1 and miR30d with chemosensitivity and prognosis of patients with epithelial ovarian cancer. Methods 76 patients with epithelial ovarian cancer underwent surgical treatment from February 2012 to January 2014 were selected as the epithelial ovarian cancer group, and 76 patients with benign ovarian tumors in our hospital were selected as the control group. The relative expressions of LncRNA PVT1 and miR30d were detected by qRTPCR. According to the sensitivity to Cisplatin (DDP), Adriamycin (ADM), Cyclophosphamide (CTX), the patients were divided into sensitive group and resistant group, and the expressions of LncRNA PVT1 and miR30d were compared. Pearson method was used to analyze the correlation between the expressions of MV30d and LncRNA PVT1. KaplanMeier was used to analyze the 5year survival of patients with epithelial ovarian cancer. Results Compared with the adjacent tissue group, the expression of miR30d in epithelial ovarian cancer group decreased (P<0.05), while the expression of LncRNAPVT1 increased (P<0.05). The expressions of miR30d and LncRNAPVT1 were closely related to FIGO stage and differentiation degree. The expression level of miR30d in DDP and ADM sensitive group was significantly higher than that in drug resistant group, and the expression level of LncRNAPVT1 was significantly lower than that in drug resistant group (P<0.05). Pearson correlation analysis showed that the expressions of MV30d and LncRNA PVT1 were negatively correlated in epithelial ovarian cancer (P<0.05). KaplanMeier analysis showed that the 5year overall survival rate and median survival time of the miR30d highexpressed group were significantly higher than those of the lowexpressed group (73.7% VS 33.3%, 2810months VS 2010months, P<0.05). The 5year overall survival rate and median survival time of LncRNA PVT1 lowexpressed group were much higher than those of highexpressed group (81.5% VS 22.4%, 2710months VS 1810months, P<0.05). Conclusion In cancer tissues of patients with epithelial ovarian cancer, the expression of miR30d is low and LncRNA PVT1 is high. The expressions of miR30d and LncRNAPVT1 are related to the chemosensitivity and prognosis of epithelial ovarian cancer.
