Abstract:【Abstract】 Objective To measure the influence of CEACAM1 in expression of inflammatory factors, cell migration and proliferation of HBE cells, and discuss the role of CEACAM1 in bronchial epithelial cells under inflammatory environment. Methods The expression of inflammatory factors including interleukin6 (IL6), interleukin8 (IL8), transforming growth factorβ (TGFβ), vascular endothelial growth factor (VEGF) in HBE cells treated with IFNγ were measured by Realtime PCR, and the upregulated factors were selected. The siRNA interfered the expression of CEACAM1 in HBE cells to observe the role of CEACAM1 in upregulation of inflammatory factors. In HBE cells were transfected with siRNACEACAM1. We also measured cell migration speed through wound healing assay, and measured cell proliferation rate by using Cell Counting Kit8 (CCK8). Results After the stimulation by IFNγ for 24 hours, the expression levels of IL6 and IL8 were up regulated in HBE cells. No significant change of TGFβ or VEGF expression was detected, and no expression of MCP1 could be measured in HBE by Realtime PCR. As compared with the negative control group, the expression levels of both IL6 and IL8 were down regulated in HBE cells transfected with siRNACEACAM1 after IFNγ stimulation. The woundhealing assay showed a delay of cell migration in HBE cells transfected with siRNACEACAM1. The cell proliferation rate also showed a decrease in HBE cells transfected with siRNACEACAM1 according to the result of CCK8 test. Conclusion IFNγ could induce the expression of IL6 and IL8 by up regulating CEACAM1. CEACAM1 contributed to the cell migration and proliferation of HBE cells.
