Abstract:【Abstract】Objective To investigate the effects of long noncoding RNA HOX transcript antisense RNA (HOTAIR) on proliferation, migration and invasion of endometrial carcinoma cells. Methods Quantitative reverse transcription PCR (qRTPCR) was used to detect the expression of HOTAIR in 20 cases of endometrial cancer and adjacent tissues. The effects of HOTAIR on proliferation, migration and invasion of endometrial cancer Ishikawa cells were studied by MTT and transwell experiments. Results The expression level of HOTAIR in endometrial cancer tissues was higher than that in adjacent tissues. At 36 h after transfection with HOTAIR siRNA, expression of HOTAIR was decreased in Ishikawa cells. At 36 h after transfection with pcDNA3.1 HOTAIR, expression of HOTAIR was increased in Ishikawa cells. The results of MTT experiments showed that inhibition of HOTAIR expression could decrease the proliferation ability of Ishikawa cells, and HOTAIR overexpression could enhance the proliferation ability of Ishikawa cells. The results of transwell experiments demonstrated that downregulation of HOTAIR expression could inhibit the migration and invasion of Ishikawa cells, and HOTAIR overexpression could promote the migration and invasion of Ishikawa cells. Conclusion LncRNA HOTAIR is highly expressed in endometrial cancer. Down regulation of HOTAIR could inhibit the proliferation, migration and invasion of endometrial cancer Ishikawa cells.
